Derivatization and Spectrophotometric Quantification of Pamidronate in Bulk and Dosage Forms | Journ

Aims: To measure pamidronate either in bulk or dosage forms in a sensitive and reliable manner.


Methodology: It is a difficult task to measure this category of drugs as they lack the involvement of chromophore groups in their structure. The proposed method relies on the derivatization of the studied drug by its 4-chloro-7-nitro-2,1,3-benzoxazole reaction and the spectrophotometric measurement of the product is 470 nm. The reaction conditions are configured with respect to the reagent length, the optimum pH for the completion of the reaction, the volume of the buffer, the optimum reaction temperature and the optimum heating period.


Results: After optimizing the reaction conditions, the studied drug can be calculated in the 9-30 μg/mL range. Validation of the system is carried out according to ICH guidelines and various validation parameters are measured and found to be excellent, such as linearity, precision, accuracy and robustness.


Conclusion: The approach proposed is precise, sensitive and can be used in quality control laboratories for the routine analysis of pamidronate.
