Determination of pKa and Degradation Products of the Mutated BRAF Inhibitor Vemurafenib and UPLC ...

The dependence of the retention factor on the pH of the mobile process was used to assess the pKa value of vemurafenib in this analysis. Using an LC-UV process, the shift in pKa in different acetonitrile fractions ranging from 57.5 to 62.5 percent (v/v) was investigated. In addition, a plain, accurate, and fast UPLC method for determining vemurafenib in human urine has been established. Internally, efavirenz was used as a standard. Selectivity factors for vemurafenib and efavirenz were found to be 1.204. For vemurafenib, the asymmetry and power factors were 1.100 and 7.532, respectively.

Thermal, photolytic, hydrolytic, and oxidative stress conditions were applied to Vemurafenib, and the stressed samples were detected using the LC/MS/MS process.
